本文研制了嵌入微柱阵列的蛇形半填充色谱柱,用于分离晚期纤维化的特征标志物。采用微机电系统( MEMS )技术制备了3 m长的矩形通道截面尺寸为250 μ m × 200μm,微柱直径为30 μ m的微分离柱。在静态涂层工艺的基础上,制备了两种不同固定相涂层的微柱( Sp1和Sp2 ),并进行了对比。详细介绍了Sp1柱只涂有聚二甲基硅氧烷( PDMS ),Sp2柱涂有氧化石墨烯量子点( GOQDs )功能化的PDMS。GOQDs是一种碳基纳米材料,具有与氧化石墨烯( GO )相似的性质,并且将微柱连接到常规气相色谱仪上进行测试。采用烷烃( C5 - C14 )和苯系物(苯、甲苯、邻二甲苯、1,3,5 -三甲苯)对微柱的分离特性进行了评价,研究了固定柱温和升温速率对色谱性能的影响。实验结果表明,在烷烃和苯系物中分别能有效分离出晚期纤维化的潜在特征标志物(戊烷和苯),且Sp2的分离效果优于Sp1。此外,Sp2柱也显示出良好的重复性。因此,GOQDs功能化PDMS涂层柱可以辅助正在开发的微系统的微检测器快速、准确、有效地区分标记物,有望成为微气相色谱( μGC )柱进行微系统集成的候选。

In this paper, the serpentine-shaped semi-packed chromatographic columns embedded with the micro-post array were developed to separate the characteristic markers of advanced fibrosis. The 3 m long micro separation columns with a rectangular channel cross-sectional dimension of 250 μm × 200 μm and the micropost diameter of 30 μm were prepared by the micro-electromechanical system (MEMS) technology. Based on the static coating process, two kinds of microcolumns (Sp1 and Sp2) with different stationary phase coatings were prepared for comparison. In detail, the Sp1 column was only coated with polydimethylsiloxane (PDMS), and the Sp2 column was coated with PDMS functionalized by graphene oxide quantum dots (GOQDs). GOQDs is a carbon-based nanomaterial that had similar properties to graphene oxide (GO). Moreover, the microcolumns are connected to a conventional gas chromatograph for testing. Alkanes (C\u003cSUB\u003e5\u003c/SUB\u003e–C\u003cSUB\u003e14\u003c/SUB\u003e) and benzene series (benzene, toluene, o-xylene, 1,3,5- trimethylbenzene) were used to evaluate the separation characteristics of microcolumns, and the effect of constant column temperature and heating rate on chromatographic performance was studied. The experimental results show that the potential characteristic markers of advanced fibrosis (pentane and benzene) could be effectively separated in alkanes and benzene series, respectively, and the separation effect of Sp2 was better than that of Sp1. In addition, the Sp2 column also showed good repeatability. Therefore, the GOQDs-functionalized PDMS coated column could assist the microdetector of the microsystem under development to rapidly, accurately and effectively distinguish the markers, and is expected to become a candidate for micro gas chromatography (μGC) column for micro-system integration.r
